I handle my corn snake once a day for about 30 minutes, is that amount of time ok once a day? Not including when she's fed and left for 48 hours.

That's fine... as long as it's not stressing them. I usually only handle about 10mins a day. Anymore and they tend to want to jump back in the viv.
